Abstract MoBi® is a systems biology software tool for multiscale physiological modeling and simulation.

It can be used for the mechanistic and dynamic modeling of biological processes and drug action.

MoBi is completely integrated with PK-Sim® [the manufacturer's package for physiologically-based pharmacokinetics (PBPK) modeling (see G6G Abstract Number 20460)] and is, consequently, especially suited for the analysis of complex pharmacokinetic and pharmacodynamic (PK/PD) models under simultaneous consideration of whole-body physiology

.

Modeling with MoBi is a unique way to integrate 'in vitro' and 'in vivo' findings into one unified representation allowing for the evaluation, analysis, interpretation and prediction of experiments to an utmost level of detail.

MoBi facilitates the process of model building and simulation of 'biochemical reaction networks' with its intuitive graphical user interface (GUI), the import and export to other modeling languages (e.g. SCAMP) and an advanced visualization tool for simulation results.

The integration of MoBi and MATLAB®, particularly the recently developed MoBi Toolbox for MATLAB (see below...), allows for the seamless performance of complex optimizations and sensitivity analyses as well as the implementation of large, highly sophisticated simulation and analysis tasks that can be programmed to run in an automated way.

MoBi's main features/capabilities are:

1) Integrated PK/PD simulations comprising both whole-body physiology and molecular mechanisms;

2) PK and PD modeling and simulation with an intuitive graphical user interface;

3) Import of PBPK models generated with PK-Sim® (PK-Sim PBPK Models);

4) Execution of simulations and investigation of simulation results by means of an advanced visualization tool;

5) Combination of multiple PK-Sim® PBPK Models;

6) Integration of PK-Sim PBPK Models with drug action models;

7) Import / Export to other modeling languages; and

8) Interface to MATLAB.

MoBi Toolbox for MATLAB --

Integrating Multiscale Physiological Modeling and Simulation with Technical Computing -

The MoBi Toolbox for MATLAB is a collection of MATLAB functions, which allow the processing of models developed in MATLAB.

For example, the MATLAB environment can be used to change parameters in a model developed in MoBi, simulate the model, and analyze the results.

This allows for an efficient operation in the model analysis stage, using the programming options as well as the function library available within MATLAB in combination with the advanced modeling interface and solver kernel included in MoBi.

In addition, the Toolbox offers efficient analysis methods tailored towards the needs of 'systems biology' and PBPK modeling including parameter identification and optimization among others.

Basic functionalities such as ‘parameter identification’ can be done without any MATLAB® or programming knowledge through Graphical User Interfaces (GUIs).

The MoBi Toolbox also provides an interface to an easy to use programming environment, which allows individual extensions to be quickly implemented.

This allows the automation of tasks and the establishment of protocols and standards.

These can vary in their level of sophistication from simple batch simulations to large-scale simulations, followed by an automated analysis including data and graphic processing.

For example, the manufacturer has developed a protocol for studying pediatric populations, which they routinely employ in projects.

Starting from an adult PBPK model that has been developed within PK- Sim (and MoBi), a set of GUIs guide you through the few project-specific specifications needed, allowing an evaluation of settings, as well as the selection of output options of interest.

The rest is executed in an automated fashion:

a) Thousands of virtual individuals are simulated and analyzed;

b) The results are visualized and written into graphical files and data files including meaningful table and figure captions; and

c) Macros within Microsoft Word® read and format the info, generating the basis for reporting to your customers or agencies.
